If mod_rewrite is not enabled then I will get a 404 error when accessing url without appending .php. I am sure that mod_rewrite engine is on our system.

Sendy won't run without mysqli. You need to request your host to install it for php.
mysqli
For 404 error, search for '404', the first result is your answer.
